双特异性嵌合抗体的制备及斑点红细胞免疫技术的建立

摘要 目的 建立一种简便、直观的检测血吸虫循环抗原的方法。方法 以N 琥珀酰亚胺基 3 (2 吡啶基二硫 )丙酸酯为交联剂制备了抗血吸虫单克隆抗体与抗人“B”型红细胞单克隆抗体的双特异性嵌合抗体 ,并建立了以人“B”型红细胞为标志物的斑点红细胞免疫技术 (Dot RCIA)用以检测血吸虫抗原。结果 初步应用于检测血吸虫循环抗原表明 ,本方法具有较好的检测效果 ,同时方法简单、可肉眼判读结果。 Objective To establish one simple,cheap,intuitional method to detect circulating antigens of Schistosoma japonicum. Methods Conjugation of anti schistosoma monoclonal antibodies and anti-human 'B' red blood cell monoclonal antibodies were prepared by means of N succinimidyl3 (2 pyridyldithio) propionate (SPDP). The Dot red blood cell immunological assay(Dot RCIA) using human 'B' red blood cell as indicator was established. Results Using Dot RCIA to detect schistosoma antigens and sera of schistosomiasis showed that this method could detect schistosoma antigens effectively. Meanwhile, the method possessed simple process,cheap cost and could determine results by naked eye. Conclusion This method has potential in detecting circulating antigens of schistosomaiasis.
