摘要 目的 探讨TUNEL法检测细胞凋亡的最佳条件。方法 各取 2 0例大肠癌组织和鼠脑组织 ,分成A、B两组 ,A组按照宝灵曼公司推荐的条件操作 ,B组按照本实验室改进后的条件操作。结果 A组与B组细胞凋亡阳性率无区别 (P >0 .0 5 ) ,A组背景着色明显高于B组 (P <0 .0 1)。 Objective To investigate the optimal conditions on apoptotic cells detected by terminal transferase deoxyuridine nick\|end labeling (TUNEL). Methods 20 human large intestinal carcinomas and 20 rat cerebrum tissues were seprated into two groups. Group A was constituted of 10 human large intestinal carcinomas and 10 rat cerebrum tissues, the rest served as group B. Group A was treated according to conditions recommended by Boehringer Mannheim company, group B was treated according to our laboratory improved conditions.Results No significant differences of positive rate of apoptotic cells were observed between group A and group B ( P >0.05).Background staining was significantly higher in group A compared to group B( P <0.01).Conclusion Improved TUNEL method could detect apoptotic cells more correctly.
