摘要
【目的】研究不同浓度黄芪注射液对米非司酮诱导滋养细胞凋亡的影响。【方法】采用体外培养滋养细胞的方法,用60μmol/L米非司酮培养滋养细胞24、48 h后撤药,加入体积分数1%、0.1%、0.01%、0.001%黄芪注射液(分别含黄芪原药材20、2、0.2、0.02 mg/L),并检测各组细胞增殖、凋亡及凋亡蛋白Caspase-3表达情况。【结果】黄芪注射液对滋养细胞增殖率无显著改善(P>0.05),但可以显著降低细胞早期凋亡率(P<0.05或P<0.01),不同程度抑制Caspase-3蛋白表达。【结论】黄芪注射液对米非司酮诱导的滋养细胞凋亡具有一定的抑制作用。
Objective To study the effects of Radix Astragali Injection (RAI) at different concentrations on mifepristone-induced apoptosis of human trophoblastic ceils in vitro. Methods The cultured trophoblastic cellswere induced by 60 μmol/L mifepristone for 24 and 48 hours, and then were treated with RAI at the volume fraction of 1%, 0.1%, 0.01% and 0.001%, which contained RA 20, 2, 022, 0.02 mg/L The proliferation of human trophoblastic cells was detected with methyl thiazolyl tetrazolium (MTT) assay, and the apoptosis was detected with Hochest33258 dye and AnnexinV-PI by flow cytometry. The expression of caspase-3 was detected with Western blot ring method. Results RAI had no effect on the proliferation of human trophoblastic cells (P〉0.05), but had an effect on markedly decreasing the early apoptotie rate of human trophoblastie cells (P〈0.05 or P〈0.01) and on inhibiting easpase-3 protein expression to various degrees. Conclusion RAI shows certain inhibitory effect on the mifepristone-induced apoptosis of human trophoblastic cells in vitro.
