Extraction and Determination of Molecular Weight Constitution of Dioscorea alata Glycoprotein

摘要 研究了水提-醇沉法提取紫山药糖蛋白。用正交设计试验考察提取温度、料液比、提取时间对提取率的影响。并分别用DEAE-52离子交换层析柱和Sephadex G-75葡聚糖凝胶柱对粗提物纯化。结果表明,提取条件为温度60℃,料液比1∶20(g/mL),提取时间3 h。粗提物经DEAE-52,Sephadex G-75柱层析纯化后得到1个糖蛋白组分,该糖蛋白经SDS-PAGE电泳分析,测得分子量为1.35×104 Da。 The extraction of glycopmtein from Dioscorea alata L. was studied. The orthogonal experiment design was applied to analyze the influence of the factors such as extracting temperature, time, ratio of solid/water on yield. The results indicated that the optimum conditions were temperature 60 ℃, time 3 h, ratio of solid/water 1 : 20 (g/mL). The coarse extractor was purified by chromatograph on DEAE-52 ion-change and Sephadex G-75 gelfiltmtion column. As a result, one kinds of glyeoprotein was collected, the relative molecular weight of the glycopmtein was 1.35× 10^4 Da with SDS-PAGE experiment.
