摘要
为研究超声波对啤酒废酵母中超氧化物歧化酶(SOD)的辅助提取效果,以总酶活和比酶活作为指标研究液料比、超声功率、超声间歇时间、超声总时间对破壁效果的影响。在单因素试验的基础上进行3因素3水平的BoxBehnken试验设计,回归模型预测的最优参数为液料比3.7︰1(mL/g)、间歇时间5 min、超声总时间4.5 min,验证值7 551.7 U和预测值7 550.9 U非常接近,回归系数R2=0.976 7,说明试验得到的回归模型能较好的应用于超声辅助提取酵母中SOD工艺参数和总酶活的预测。
An ultrasonic-assisted extraction process for separation of superoxide dismutase (SOD) from beer yeast mud was developed. Effects of liquid to solid ratio, ultrasonic power, ultrasonic time interval and treating time on the total enzyme activity and specific enzyme activity were studied. The liquid to solid ratio, ultrasonic time interval and treating time were further studied using a Box-Behnken design to optimize the extract technology. Results of the response surface optimization test showed that the optimal extract condition was as following: liquid to solid ratio 3.7 : 1 (mL/g), ultrasonic time interval 5 s, treating time 4.5 min. The predicated average TEA was 7 550.9 U with the optimal extract condition, and the verification tests demonstrated that the actual value was 7 551.7 U, R2 was 0.976 7. The results indicated that the extract system model had an extremely significant effect on oredictinz the result of the test.
