Determination of Eleven Polyphenolic Compounds in Mulberry Wine by High Performance Liquid Chromatography

摘要 建立了高效液相色谱同时测定桑葚酒中11种多酚类化合物的方法。采用Venusil C18色谱柱(250 mm×4.6mm,φ5μm)分离11种酚类物质,流动相为50%乙腈和超纯水,分别加1%乙酸,进样量20μL,采用梯度洗脱,流速为1.0 mL/min,柱温30℃,检测波长260 nm、320 nm。各组分的质量浓度与其峰面积具有良好的线性关系,相关系数均大于0.999 6,且11种酚类物质在45 min内得到了较好分离。方法回收率在80.3%~105.6%之间,相对标准偏差为1.04%~2.50%,检出限为0.02~0.05 mg/L。该方法可用于桑葚酒中多酚类化合物的快速分析。 A method was developed for the simultaneous determination of eleven polyphenolic compounds in mulberry wine, they were separated by HPLC on a Venusil C18 (250 mm ×4.6 mm, φ5 μm) column 30 ℃, gradient elution was employed with a moble phase consisting of 50% acetonitrile and water, and add to 1% acetic acid, a solvent flow rate of 1.0 mL/min and an injection volume of 20 μL. Eleven polyphenolic compounds were identified at 260 nm and 320 nm. The method showed good linear correlations between the concentration and areas of the eleven components with correlation coefficients greater than 0.999 6, and were separated well in 45 min. The average recoveries were 80.3%-105.6%, with relative standard deviations of 1.04%-2.50%, and the LOD is between 0.02-0.05 mg/L. The method can be applied to the rapid determination ofpolyphenolic compounds in mulberry wine.
