丹参酮ⅡA联合参麦注射液对肾缺血-再灌注大鼠氧化应激及肾功能的影响 被引量:8

摘要 目的探讨丹参酮ⅡA联合参麦注射液对大鼠肾缺血-再灌注(ischemia-resperfusion,IR)肾组织中氧化应激水平及肾功能的影响。方法 200只SD大鼠随机分为假手术组、IR模型组、丹参酮ⅡA组、参麦组和丹参酮ⅡA联合参麦组,分别予以干预。测定各组大鼠肾组织中丙二醛(malonaldehyde,MDA)和超氧化物歧化酶(superoxide dismutase,SOD)含量的变化,同时检测血肌酐(creatinine,Cr)和尿素氮(blood urea nitrogen,BUN)水平。结果 IR模型组肾组织匀浆MDA水平随再灌注时间的延长逐渐升高,12 h达高峰后下降,在6 h和12 h与其它组相比较有统计学意义(P<0.05),而丹参酮ⅡA组、参麦组、丹参酮ⅡA联合参麦组和假手术组中MDA水平无此变化,且4组相互比较差异无统计学意义(P>0.05)。丹参酮ⅡA组、参麦组在24 h、丹参酮ⅡA联合参麦组在12 h和24 h与IR组比较不同程度地提高SOD的活力(P<0.05),与假手术组比较差异无统计学意义(P>0.05);丹参酮ⅡA联合参麦组与丹参酮ⅡA组、参麦组比较在24 h SOD活力提高(P<0.05);丹参酮ⅡA组与参麦组比较差异无统计学意义(P>0.05)。丹参酮ⅡA组、参麦组和丹参酮ⅡA联合参麦组血Cr和BUN水平低于IR组(P<0.05);丹参酮ⅡA联合参麦组与丹参酮ⅡA组和参麦组比较,在24h血Cr和BUN值均降低明显,差异有统计学意义(P<0.05)。结论丹参酮ⅡA联合参麦注射液通过抗氧自由基损伤及减轻脂质过氧化反应保护肾功能。 Objective To investigate the effect of tanshinoneⅡ A and shenmai injection on oxidative stress and renal function in rats after ischemia-reperfusion(IR) injury.Methods Two hundred SD rats were randomly divided into sham operation group(SOG),IR injury model group(IRG),tanshinoneⅡ A group(TG),shenmai injection group(SIG),and tanshinoneⅡ A + shenmai injection group(TSG).Kidney tissue levels of malonaldehyde(MDA) and superoxide dismutase(SOD) were measured.At the same time,serum creatinine(Scr) and blood urea nitrogen(BUN) were also determined.Results in the IRG,the MDA levels in renal tissue homogenate were gradually increased with the prolongation of reperfusion,reached peak at 12 hours,and then decreased.The levels at 6 h and 12 h were significantly different from the other groups(P 〈 0.05).No significant difference was found in the MDA levels among the TSG,SIG,TSG and SOG.The SOD activity in the TG and SIG at 24 h,and in the TSG at 12 h and 24 h after IR injury was higher than that in the IRG(P〈0.05) but there was no significant difference when compared with the SOG.Moreover,the SOD activity was higher in the TSG than that in the TG or SIG at 24 h after IR injury(P〈0.05).The serum Scr and BUN levels were lower in the TG,SIG and TSG than that in the IRG,and lower in the TSG than in the TG or SIG at 24 h after IR injury(P〈0.05).Conclusion TanshinoneⅡ A and shenmai injection can protect renal function by reducing its anti-free radical injury and alleviating its lipid peroxidation.
