摘要 过氧化物酶体增生激活受体γ(PPARγ)协同刺激因子-1α(PGC-1α)是近年来发现的一种核转录辅激活因子,能通过与核受体和转录因子相互作用调节线粒体氧化代谢过程,调控线粒体生物合成,维持线粒体结构与功能的完整性;调节能量代谢过程中的基因表达、调控能量与营养平衡.近期研究显示它与代谢疾病、病毒感染、炎症反应、肿瘤的发生、发展密切相关.通过调节PGC-1α生理功能来治疗疾病和研发药物已成为研究热点. The peroxisome proliferator-activated receptor (PPAR) γ coactivator 1-α (PGC-1α) is a transcriptional coactivator that regulates many aspects of oxidative metabolism involving in mitochondrial biogenesis and the maintaince of the integrity of mitochondrial structure through the coactivation of many nuclear receptors and transcriptional factors.PGC-1 α may play a role in the regulation of gene expression for energetic metabolism and the homeostasis of energy and nutrition.Recent studies on abnormal expression of PGC-1α have revealed significant association between this molecular and the clinical disorders,such as metabolic diseases,tumors,viral infection and inflammation.The biological activities of PGC-1 α and the association with the potential diseases has become the new research focus and this strategy could be a novel target for drug development.
