血府逐瘀汤对大鼠局灶性脑缺血损伤的保护作用及其作用机制 被引量:23

Protective effects of Xuefu Zhuyu decoction on focal crerebral ischemic injury in rats and its mechanism of actions

摘要 目的:探讨血府逐瘀汤对局灶性脑缺血损伤大鼠的保护作用及其可能的作用机制。方法:采用大鼠永久性大脑中动脉阻塞(MCAO)方法建立局灶性脑缺血模型,灌胃给予血府逐瘀汤(5,10,20 g·kg-1)进行干预,通过神经症状评分评价大鼠行为学的改变,TTC染色法观察脑梗死范围,紫外分光光度法检测缺血脑组织丙二醛(MDA)、一氧化氮(NO)含量及超氧化物歧化酶(SOD)活性,实时荧光定量PCR技术检测缺血大脑皮层凋亡相关基因Bax、Bcl-2、Caspase-3的mRNA表达。结果:血府逐瘀汤可显著改善MCAO大鼠神经症状,减少脑梗死体积,降低缺血脑组织中MDA、NO含量,提高SOD活性,同时下调缺血大脑皮层Bax、Caspase-3的mRNA表达,上调Bcl-2 mRNA表达。结论:血府逐瘀汤对局灶性脑缺血大鼠脑损伤有保护作用,其作用机制可能与其抗氧化及抑制凋亡有关。 OBJECTIVE To investigate the protective effects of Xuefu Zhuyu decoction on focal crerebral isehemic injury in rats and its possible mechanism of actions. METHODS The method of permanent middle cerebral artery occlusion in rats (MCAO) was adopted to establish focal cerebral ischemia model, MCAO rats were given Xuefu Zhuyu decoction (5, 10, 20 g· kg-1) through the intragastric administration. Evaluation index like cerebral infarction volume and behavioral symptoms of brain were taken account in terms of Xuefu Zhuyu decoction in cerebral ischemia neural protection. The concentrations of ma- londialdehyde (MDA) and nitric oxide (NO), activities of superoxide dismutase (SOD) in cerebral tissue were measured by UV speetrophotometry. Real time PCR was applied to detect Bax, Bcl-2 and Caspase-3 mRNA expression on ischemic cerebral cor- te~ RESULTS In rat focal cerebral ischemia model, Xuefu Zhuyu decoction could significantly reduce the infarction volume and improve brain behavior signs. The concentrations of MDA and NO in brain tissue were lower while activities of SOD were higher. Real time PCR results showed.- Bax and Caspase-3 mRNA expression in MCAO rat cortex isehemia could down-regula- ted and the levels of Bcl-2 up-regulated. CONCLUSION Xuefu Zhuyu decoction has protective effects on cerebral ischemia inju- ry in rats, which is probably related to antioxidant and the inhibition of apoptosis.
