摘要
目的研究制粒因素对淫羊藿总黄酮酶解的影响。方法将蜗牛酶、淫羊藿总黄酮和糊精混合经湿法制粒制成含酶淫羊藿总黄酮颗粒,HPLC法测定其酶解过程中淫羊藿苷、朝藿定A、朝藿定B、和朝藿定C以及酶解产物(宝藿苷Ⅰ、箭藿苷A、箭藿苷B、鼠李糖基淫羊藿次苷Ⅱ和淫羊藿苷元)的含有量变化。结果与未经制粒的淫羊藿总黄酮粉末酶解相比,经糊精湿法制粒得到的含酶淫羊藿总黄酮颗粒在37℃缓冲溶液中酶解时淫羊藿苷、朝藿定A、朝藿定B和朝藿定C的酶解速率变慢,但在肠道排空时间(4~6 h)内可以完全酶解转化成次级苷宝藿苷Ⅰ、箭藿苷A、箭藿苷B和鼠李糖基淫羊藿次苷Ⅱ,次级苷继续被水解可生成淫羊藿苷元。结论糊精湿法制粒对淫羊藿总黄酮的酶解转化有一定影响。
AIM To investigate the effect of dextrin wet granulation on enzymolysis of total flavonoids from epimedium. METHODS To mix dextrin with snailase and total epimedium flavonoids to granulate the sample.HPLC was used to measure the content changes of the main flavonoids,including icariin,epimedin A,epimedin B,and epimedin C; and enzymolysis products baohuosideⅠ,sagittatoside A,sagittatoside B,2″-O- rhamnosylicarisideⅡand icaritin. RESULTS As compared with non-wet granulation,hydrolysis rates of icariin,epimedin A,epimedin B and epimedin C slowed down in buffer solution( 37 ℃),but all were completely degraded into baohuosideⅠ,sagittatoside A,sagittatoside B and 2″-O-rhamnosylicariside Ⅱat the intestinal evacuation intervals( 4 ~6 h). Meanwhile the secondary glycoside kept on its hydrolyzing into aglycone. CONCLUSION Dextrin wet granulation has some effects on the enzymolysis of total flavonoids from epimedium.
