四神丸对溃疡性结肠炎大鼠血清TLR4、TGF-β1及IL-10的影响

Effects of Sishen Pill on Serum TLR4,TGF-β1 and IL-10 in Ulcerative Colitis Rats

目的:探讨四神丸治疗溃疡性结肠炎(UC)的作用机制。方法:将40只SD大鼠随机分为正常组、模型组、对照组及四神丸组各10只。除正常组外,其余各组均采用2、4、6-三硝基苯磺酸(TNBS)/乙醇法复制UC大鼠模型,灌胃14 d后观察各组大鼠血清TLR4、TGF-β1、IL-10的变化。结果:模型组大鼠血清TLR4水平增高,TGF-β1、IL-10水平降低,与正常组比较,差异均有统计学意义(P<0.01);四神丸组TLR4、TGF-β1、IL-10均较模型组明显改善(P<0.05或P<0.01)。结论:四神丸可调节溃疡性结肠炎大鼠TLR4-DC免疫耐受失调,可能是其治疗UC的作用机制之一。

Objective To explore the mechanism of action of Sishen Pill （SP） on ulcerative colitis （UC） rats. Methods：The 40 rats were randomly divided into normal group, model group, control group and SP group, 10 rats in each. Except nor-mal group, the others were used 2,4,6-trinitrobenzene sulfonic acid （TNBS）/ethanol method copied UC rat model. The changesof serum TLR4, TGF-β1, IL-1 was observed in each group after gavage 14d. Results： The levels of serum TLR4 increased,TGF-β1, IL-10 decreased significantly in model group, compared with the normal group （P〈0.01）. The TLR4, TGF-131 andIL-10 significantly improved in SP group, compared with model group （P〈0.05 or P〈0.01）. Conclusion： Sishen pill can regulatethe immune tolerance of TLR4-DC dysregulation, which may be the mechanism for the treatment of UC. [
