血府逐瘀汤对心肌缺血大鼠模型血清NO及缺血区心肌组织VEGF、DLL4蛋白表达的影响 被引量:16

Effects of Xuefu Zhuyu Decoction on the Expression of VEGF, DLL4 Protein and the Content of NO in rats with Myocardial Ischemia

摘要 目的:探讨血府逐瘀汤对心肌缺血大鼠模型缺血区心肌血管新生的影响及其祛瘀生新(生新络)作用机制。方法:采用结扎大鼠冠状动脉左前降支制备急性心肌缺血模型,用血府逐瘀汤进行干预,分别于第7天、第14天处死动物取血,采用硝酸还原酶法检测大鼠血清NO的含量,HE染色观察缺血区心肌形态学的改变,免疫组化法检测缺血区心肌血管内皮细胞生长因子(VEGF)、Delta样配体4(Delta-like ligand 4,DLL4)蛋白的表达。结果:与假手术组比较,模型组血清NO含量明显降低(P<0.01),缺血区心肌VEGF、DLL4蛋白表达增加(P<0.05);与模型组比较,血府逐瘀汤组、麝香保心丸组血清NO含量均升高(P<0.05),缺血区心肌VEGF、DLL4蛋白表达明显增加(P<0.01)。结论:血府逐瘀汤可促进心肌缺血大鼠模型缺血区心肌血管新生,对实验性心肌缺血损伤有保护作用,其祛瘀生新(生新络)作用与提高血清NO含量及缺血区VEGF、DLL4蛋白表达有关。 Objective To investigate the effect of Xuefu zhuyu Decoction on angiogenesis in ischemic myocardium in ratswith myocardial ischemia and the mechanism of removing blood stasis for promoting tissue regeneration. Methods: The ligation ofleft anterior descending coronary artery was applied to establish rat acute myocardial ischemia model, with intervention of XuefuZhuyu Decoction. The rats were killed in 7 th and 14 th day respectively for blood. NO in serum was detected by nitrate re-ductase method. HE staining was used to observe the morphological changes of ischemic cardiac muscle; Immunohistochemistrywas used to detect the myocardial ischemic vascular endothelial cell growth factor (VEGF), Delta like ligand 4 (Delta-like ligand4, DLIA) protein expressions. Results: Compared with the sham operation group, the content of NO in model group decreasedsignificantly (P〈0.01), and VEGF and DLIA protein expression increased (P〈0.05); Compared with the model group, the contentof NO in serum were increased (P〈0.05) in Xuefu Zhuyu Decoction group and Shexiang Baoxin Pill group, and the expressionof VEGF and DLIA protein in ischemic myocardium was significantly increased (P〈0.01) in Xuefu Zhuyu Decoction group andShexiang Baoxin Pill group. Conclusion: Xuefu Zhuyu Decoction can promote angiogenesis in isehemic myocardium in rats withmyocardial ischemia. The mechanism of removing blood stasis for promoting tissue regeneration is related to the increase thecontent of NO in serum and protein expression of VEGF and DLIA in ischemia myocardium.
