摘要 目的观察miR-19b对巨噬细胞ABCA1表达的影响。方法在THP-1源性巨噬细胞中转染miR-19b模拟物(mimic)和抑制物(inhibitor),荧光定量PCR检测ABCA1 mRNA,Western blot检测ABCA1蛋白水平。结果转染miR-19b mimic后,巨噬细胞ABCA1 mRNA及蛋白水平均显著下调;转染miR-19b inhibitor后ABCA1蛋白则明显上调,但ABCA1 mRNA无变化。结论 miR-19b下调巨噬细胞ABCA1的表达。 Objective To investigate the impact of miR-19b on the expression of macrophage ABCA1. Methods The protein and mRNA levels of ABCA1 were detected by Western blot and quantitative real time PCR( qRT-PCR) respectively in THP-1 macrophage transfected with miR-19b mimic and inhibitor. Results The protein and mRNA levels of ABCA1 were obviously downregulated in THP-1 macrophage transfected with miR-19b mimic. The protein level of ABCA1was evidently upregulated in THP-1 macrophage transfected with miR-19b inhibitor,while ABCA1 mRNA level had no significantly alternation. Conclusion miR-19b regulates the expression of macrophage ABCA1.
