辐照结合束缚应激致血虚肝郁大鼠证候模型的建立 被引量:12

Establishment of rat model of blood deficiency and liver depression syndrome induced by the method of chronic restraint stress combined with radiation

摘要 目的:建立辐照结合束缚应激致血虚肝郁证大鼠模型。方法:SD雄性大鼠随机分为空白组、辐照组、束缚组、模型组,模型组采用大鼠单笼饲养、辐照结合束缚21d造模,检测各组大鼠体质量、外周血象并进行糖水实验、敞箱实验和跳台实验,测定血浆中促肾上腺皮质激素(ACTH)、皮质酮(CORT),下丘脑促肾上腺皮质激素释放激素(CRH)和大脑海马单胺类神经递质5-羟色胺(5-HT)和多巴胺(DA)等含量。结果:模型组大鼠生长缓慢,外周白细胞,红细胞、血红蛋白下降明显(P<0.05),血浆中ACTH明显升高(P<0.05),CORT和下丘脑CRH含量有升高趋势,脑内单胺类神经递质5-HT和DA含量明显降低(P<0.05)。结论:从血象、行为学、下丘脑-垂体-肾上腺(HPA)轴和脑内单胺类神经递质等方面验证了辐照结合束缚应激造成的大鼠模型可作为中医血虚肝郁证的动物模型。 Objective: To establish the rat model of blood deficiency and liver depression syndrome induced by the method of chronic restraint stress combined with radiation. Methods: SD male rats were randomly divided into four groups which recorded as normal group, radiation group, chronic restraint stress group and model group. The rats in the model group were prepared by chronic restraint stress combined with radiation respectively for 21 days and were fed separately. The sucrose preference test, openfield test and step down test were carried out, and the weight, peripheral hemogram, ACTH and CORT in plasma, CRH in hypothalamus, 5-HT and DA in hippocampus were measured. Results: The rats in the model group grew slowly, and the white blood cells, red blood cells and hemoglobin in peripheral blood decreased signii cantly(P〈0.05). The ACTH in plasma increased significantly(P〈0.05). CORT, CRH in hypothalamus increased, and 5-HT and DA decreased(P〈0.05). Conclusion: It verii es from hemogram, ethology, HPA axis and the content of monoamine neurotransmitters that the rat model induced by the method of chronic restraint stress combined with radiation can be used as the model of blood deficiency and liver depression syndrome.
