摘要
目的:观察清泻浊毒法对急性痛风性关节炎模型大鼠的抗炎作用,探讨其作用机制。方法:大鼠随机分为空白组、模型组、清泻组、吲哚组。采用踝关节注射尿酸钠建立急性痛风性关节炎模型。观察大鼠关节肿胀度及其组织病理改变,测定关节组织中的白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)含量。结果:清泻组能够明显减轻大鼠关节肿胀度,造模后24h、72h分别与造模前比较均无显著性差异;清泻组与吲哚组均使升高的IL-1β、TNF-α水平降低,与模型组比较有显著性差异(P<0.05,P<0.01);两治疗组光镜下均仅有少量炎性细胞浸润,未见软骨破坏。结论:清泻浊毒法能够明显改善急性痛风性关节炎大鼠的关节肿胀,控制局部炎症,阻止软骨及软骨下骨质破坏。其作用机制与降低IL-1β、TNF-α等炎症细胞因子水平有关。
Objective: To observe and discuss the anti-inflammatory effect and mechanism of Qingxie Zhuodu method on the model rats with acute gouty arthritis. Methods: Rats were randomly divided into blank group, model group, Qingxie Zhuodu Recipe group, indometacin group. The rat model of acute gouty arthritis was set up by injection of sodium urate on ankle. The swelling degrees and pathological changes in rats joint were observed. The expressions of IL-1β and TNF-α in rats joint were measured. Results: Qingxie Zhuodu Recipe group signii cantly relieved swelling degrees of rats joint. Compared with that before model establishment, there was no signii cant dif erence after modeling for 24 and 72 hours. The IL-1β and TNF-α expression of Qingxie Zhuodu Recipe group and the indometacin group both decreased signii cantly, which were signii cant dif erences compared with those of the model group(P〈0.05, P〈0.01). The two treatment groups were observed a small amount of inflammatory cell ini ltration in light microscope, without cartilage destruction. Conclusion: Qingxie Zhuodu method can signii cantly improve the swelling degrees of rats with acute gouty arthritis, control the inl ammation around rat joints and prevent the destruction of cartilage and subchondral bone. Its mechanism might relate to the effect of decreasing the level of inflammatory cytokine such as IL-1β and TNF-α.
