摘要
目的:从不同角度观察脑脉舒康胶囊(脑脉舒康)对沙土鼠脑缺血再灌注损伤的保护作用。方法:采用结扎双侧颈总动脉缺血10min,再灌注60min,建立沙土鼠脑缺血再灌注损伤模型。观察脑脉舒康对沙土鼠脑缺血再灌注模型血浆中内皮素(ET)和降钙素基因相关肽(CGRP)含量,以及脑组织匀浆中的血栓素(TXB2)、谷氨酸(Glu)、氨基酸(EAA)含量、ATP酶活力的影响及光镜观察脑海马神经组织及细胞形态学变化。结果:与模型组比较,脑脉舒康能显著降低沙土鼠脑缺血再灌注损伤模型血浆中ET含量(P<0.05,P<0.01),升高CGRP含量(P<0.05,P<0.01);显著降低脑组织中TXB2、Glu和EAA含量(P<0.01),提高其ATP酶活性(Na+-K+-ATP、Ca2+-ATP、Mg2+-ATP);病理观察结果显示,脑脉舒康组部分脑神经细胞得到一定的恢复。结论:脑脉舒康对沙土鼠脑缺血再灌注损伤具有一定的治疗作用,其作用机制可能和改善脑能量代谢障碍、对抗脑缺血后EAA升高及保护血管内皮细胞有关。
Objective: To observe the protective effects of Naomai Shukang Capsules on cerebral ischemia reperfusion injury in gerbils from dif erent ways. Methods: The model of cerebral ischemia reperfusion injury in gerbils was established by fastening bilateral common carotid artery for 10 minutes and reperfusing for 60 minutes. The contents of ET and CGRP in blood plasma were observed, the contents of TXB2, Glu, EAA, ATP activity in brain tissue were detected, in order to observe the effect of Naomai Shukang Capsules on cerebral ischemia reperfusion injury in gerbils, and brain morphology and hippocampal neuron were observed by using microscopy. Results: Compared with the model group, the content of ET in blood plasma was signii cantly lower(P〈0.05, P〈0.01) and the content of CGRP was higher in Naomai Shukang Capsules group(P〈0.05, P〈0.01). The contents of TXB2, Glu, EAA was decreased(P〈0.01), the ATP activity was increased in Naomai Shukang Capsules group(Na+-K+-ATP, Ca2+-ATP, Mg2+-ATP). Pathological examination showed that Naomai Shukang had signii cant protection against neuronal injury in the hippocampus. Conclusion: Naomai Shukang Capsules have therapeutic effect on improving the injury degree of brain on cerebral ischemia reperfusion injury in gerbils. The mechanism could be related to improving the brain energy metabolism, inhibiting the activity of EAA and protecting vascular endothelial cells.
