摘要
目的阐述β2-糖蛋白Ⅰ(β2-GPⅠ)在抗心磷脂抗体阳性先兆流产中诱导抗体产生的机制,为安子合剂治疗ACA阳性先兆流产的进一步研究打下良好的实验基础。方法以β2-GPⅠ为免疫原,构建流产小鼠模型;采用酶联免疫吸附法(ELISA)检测抗心磷脂抗体阳性先兆流产模型小鼠血清中ACA、抗β2-GPⅠ抗体。结果①成功复制模型:胚胎丢失率模型组明显高于空白组(P〈0.05),模型组抗β2-GPⅠ抗体浓度、ACA滴度均显著高于空白组(P〈0.05);②安子合剂低剂量组能明显降低抗β2-GPⅠ抗体浓度和ACA滴度(P〈0.01),且效果优于安子合剂中剂量组、高剂量组以及阿司匹林组(P〈0.05~0.01)。结论①抗β2-GPⅠ抗体、ACA均是导致免疫性复发流产的免疫因素;②安子合剂通过降低ACA滴度和抗β2-GPⅠ抗体浓度改善ACA阳性先兆流产妊娠结局;③安子合剂低剂量组具有临床等效性,随着剂量的增加,反而引起代谢障碍而使得效果逐渐减弱。
OBJECTIVE To investigate the mechanism ofβ2-GPⅠin ACA-positive threatened abortion,and to lay agood experimental foundation for the further study of treatment of Anzi mixture in ACA-positive threatened abortion.METHODSβ2-GPⅠ,as an immunogen,was applied to establish the abortion model;the ACA andβ2-GPⅠantibody in serum of abortion model rats were detected by enzyme linked immunosorbent assay(ELISA).RESULTS ①Successful established animal model:The embryo loss rate of model group was obviously higher than that of the blank group.The concentration ofβ2-GPⅠantibody and drop degree of ACA of model group was obviously higher than that of blank group;②The low dose group obviously decreased the concentration ofβ2-GPⅠantibody and the drop degree of ACA,which of the effect was better than the medium dose group,the high dose group and the aspirin group.CONCLUSION ①β2-GPⅠantibody and ACA are immune factors which lead to immunity recurrent miscarriage;②Anzi mixture may improve pregnancy outcome of ACA-positive threatened abortion by reducing the drop degree of ACA andβ2-GPⅠantibody concentration;③The low dose group has clinical equivalence,while the increasing of dose may lead to metabolic disorders and reduce curative effect.
