摘要
目的运用高效液相色谱法建立升麻葛根汤的指纹图谱,并考察白芍硫磺熏蒸后对升麻葛根汤指纹图谱的影响。方法采用Elite C18色谱柱(250mm×4.6mm,5μm),流动相为甲醇-0.3%甲酸水溶液,梯度洗脱;流速1.0mL/min;柱温30℃,检测波长250nm。结果通过对21批(8批含有未硫熏白芍和13批含有硫熏白芍)升麻葛根汤的HPLC指纹图谱研究,建立了升麻葛根汤HPLC指纹图谱的共有模式,确立了10个共有峰。通过相似度计算并结合系统聚类分析和主成分分析对21批升麻葛根汤的HPLC指纹图谱进行分析比较,结果显示未硫熏白芍和硫熏白芍组成的升麻葛根汤可明显分成两类。结论该方法准确可靠,重复性好,可用于升麻葛根汤的质量控制和评价。
OBJECTIVE RP-HPLC method was applied to establish the chromatographic fingerprints of Shengma Gegen Decoction,as well as the changes of HPLC fingerprints of Shengma Gegen Decoction caused by sulfur-fumigated Paeoniae Radix Alba were investigated.METHODS The separation was performed on an Elite C18column(250mm×4.6mm,5μm)using agradient elution with mobile phase composed of methanol-aqueous solution containing 0.3%(v/v)formic acid.The column temperature was 30℃ with a flow rate of 1.0mL/min.The detection wavelength was set at 250 nm.RESULTS The common patterns and ten common peaks of HPLC fingerprints of Shengma Gegen Decoction were established by analyzing twenty one batches of Shengma Gegen Decoction samples which composed of both eight batches of those contained sun-dried and thirteen batches of those contained sulfur-fumigated Paeoniae Radix Alba.The results showed that Shengma Gegen Decoction contained sun-dried Paeoniae Radix Alba and Shengma Gegen Decoction contained sulfur-fumigated Paeoniae Radix Alba could be obviously divided into two categories by comparison and evaluation using similarity algorithm combined with hierarchical cluster analysis and principal component analysis for HPLC fingerprints of twenty one batches of Shengma Gegen Decoction samples.CONCLUSION The established method is reliable with good reproducibility and can be suitable for quality control and evaluation of Shengma Gegen Decoction.
