磷脂混杂酶1在肝癌中的表达及其临床意义 被引量:1

摘要 目的 观察磷脂混杂酶1(PLSCR1)对肝癌生长转移的调控作用。方法 构建PLSCR1干扰细胞株,通过噻唑蓝(MTT)实验和侵袭(Invasion)实验分别研究PLSCR1在肝癌细胞HepG2的增殖和侵袭中的作用。通过荧光定量聚合酶链反应(FQ-PCR)检测20例肝癌患者的癌组织及癌旁组织中PLSCR1的表达,探讨PLSCR1在肝癌生长及转移中的意义。结果 成功构建PLSCR1-小干扰RNA(siRNA)-HepG2细胞株,并通过MTT实验和Invasion实验证实干扰PLSCR1对肝癌细胞的生长及侵袭具有抑制作用,其抑制率分别为(28.71±1.27)%和(27.16±10.40)%。FQ-PCR检测20例肝癌患者的癌组织及癌旁组织中PLSCR1的表达,发现PLSCR1在肝癌组织中的表达明显高于癌旁组织(ΔΔCt:-2.57±1.34),并与肿瘤分化程度和临床分期等病理因素明显相关(P〈0.05)。结论 PLSCR1在肝癌组织中高表达,并与肝癌的发生、发展密切相关。 Objective To investigate the regulatory role of phospholipid scramblase 1 (PLSCR1) during the growth and metastasis of hepatic carcinoma. Methods The PLSCR1 interference cell lines were constructed. The significance of PLSCR1 during cell proliferation and invasion was measured by methyl thiazol tetrazolium (MTT) assay and Invasion assay. The expression of PLSCR1 was detected by real-time fluorescent quantitative polymerase chain reaction(FQ-PCR) in 20 patients with hepatic carcinoma. Results The PLSCRI-small interfering RNA (siRNA)-HepG2 cell lines were successfully constructed. It was found the PLSCR1 interference could suppress the growth and invasion, respectively, the inhibition rate is separately (28.71 ± 1.27( % and (27.16 ± 10.40) %. The results of FQ-PCR indicated PLSCR1 was overexpressed in tumor tissue ( A ACt : - 2. 57 ± 1.34), and it was related with the tumor differentiation and clinical stages (P 〈 0. 05). Conclusion PLSCR1 is over-expressed in hepatic carcinoma, and has close correlation with tumor growth and progress.
