溴脱氧尿苷标记滞留细胞在SD大鼠足垫皮肤及汗腺的分布

摘要 目的:将溴脱氧尿苷(BrdU)作为标记滞留细胞(LRCs)标记物,通过对皮肤组织中LRCs的检测,证实干细胞的存在及其分布。方法:SD大鼠予50 mg·kg^(-1)·次^(-1)BrdU腹腔注射,每天4次,每次间隔2h,连续2d。最后一次注射后第6周,处死之,取双后足垫。免疫组化染色对表达BrdU细胞进行检测。结果:BrdU阳性细胞散在分布于真皮层的外泌汗腺蟠状分泌部、弯曲和直导管部。BrdU阳性细胞表达率在外泌汗腺(4.2±1.3)%)明显高于在表皮基底层(0.5±0.1)‰)(P<0.05)。结论:大鼠后足垫外泌汗腺中存在的这些干细胞或许在皮肤及其附件损伤修复中发挥重要作用。
