中药复方962胶囊对老年大鼠海马神经生长因子及其受体表达的影响 被引量:15

Effect of Chinese compound 962 capsule on expression of nerve growth factor and Trk A receptor in hippocampus of aged rats

摘要 目的 我们以往的实验证明 :中药复方 96 2胶囊 (含何首乌、石菖蒲等六味中药 )能显著改善老年大鼠学习记忆功能。本研究观察 96 2胶囊对自然衰老大鼠模型海马区神经生长因子(NGF)及其受体TrkA表达的影响 ,探讨 96 2胶囊改善学习记忆功能的作用机理。方法 雌性Wistar大鼠 ,老年为 2 2月龄 ,青年为 5月龄。 96 2胶囊高剂量 1 8g/kg ,96 2胶囊低剂量 0 9g/kg ,每日灌胃给药一次 ,连续灌胃 2个月 ,ABC法行NGF和TrkA免疫组化染色。VISILOG5 0图像分析系统对组织切片进行分析。海马CA1区锥体细胞层连续摄取 3个视野 ,这 3个视野包括了整个CA1区 ,记数 3个视野内总阳性细胞数量、阳性细胞总面积 (象素单位 )及阳性细胞灰度积分值 (灰度单位 )。海马CA2区连续摄取 2个视野 ,海马CA3区连续摄取 2个视野 ,海马CA4区摄取齿状回包含的 1个视野 ,包括了完整的CA2、CA3、CA4区。结果 ①老年大鼠海马CA1、CA2、CA3、CA4区NGF和TrkA阳性细胞数、阳性细胞总面积和阳性细胞灰度积分值较青年对照组明显减少 ,以CA2、CA3区的减少最严重。② 96 2胶囊能提高老年大鼠海马CA1、CA2、CA3、CA4区NGF和TrkA阳性细胞数量、阳性细胞总面积和阳性细胞灰度积分值 ,96 2胶囊高剂量组基本恢复青年对照组水平。结论 老年大鼠神经生? Objective In the pathology of Alzheimer's disease,age is the most important risk factor.For mimicking the pathological abnormality of the disease,we take the aged rats as the animal model of Alzheimer's disease.Our previous experiments demonstrated Chinese compound 962 capsule (composed of six kinds of Chinese herbs such as heshouwu and shichangpu) improved the learning and memory ability of aged rats significantly.In order to study the protective mechanism of this drug,we observed the expression of nerve growth factor (NGF) and its receptor (Trk A) in hippocampus of aged rats with or without 962 capsule administrated. Methods\ Aged Wistar rats(22 months old)were administrated with 962 capsule for 2 months (once a day) before testing the learning and memory ability.Positive control drug was Piracetam (0.56?g/kg),high dose of 962 capsule was 1.8?g/kg and low dose of this drug was 0.9?g/kg. After learning experiment the animals were killed for immunohistochemical analysis.Immunohistochemistry for the NGF and Trk A receptor were carried out in frozen slice (40?μm thick) of brain by ABC method according to a reported procedure.Image analysis to determine average cell number,size(area) and staining density in hippocampal CA1,CA2,CA3 and CA4 regions was achieved with VISILOG 5.0 image pattern analysor. Result\ ①Both NGF\|and TrkA\|immunoreactive neurons in aged rats significantly decreased in cell number,size and staining density in four regions of hippocampus.Decreasement in CA2 and CA3 region was most severe.among all four regions.②The significant increase in cell number,size and staining density of NGF\|and TrkA\|immunoreactive neurons in four regions of hippocampus was observed in 962 capsule administrated groups. Conclusion\ Decreased level of NGF and its receptor (Trk A) in hippocampus may account for the deficiency of learning and memory ability in aged rats.Chinese compound 962 Capsule administration significantly promotes the expression of NGF and Trk A in aged rats,and it may be the mechanism by which 962 Capsule improves the learning and memory ability.Enhancing endogenous neurotrophic substance plays an important role in the treatment of Alzheimer's disease.\;[
