黑青稞籽皮类黄酮的提取及其对缺氧损伤细胞的保护作用 被引量:13

摘要 目的探讨不同缺氧损伤细胞模型下黑青稞籽皮类黄酮物质对缺氧损伤细胞的保护作用。方法采用柠檬酸-乙醇浸提法富集黑青稞籽皮类黄酮物质,并对类黄酮含量进行两种不同方法检测和定量,用三种不同的细胞模型验证其对缺氧损伤的保护作用。结果实验条件下得到黑青稞籽皮提取物30.3 mg/g,通过光谱分析及HPLC检测发现,籽皮中的类黄酮物质多为花青素类,花青素提取率为68.6 mg/g,其中氯化花翠素占提取物的57 mg/g;以芦丁标定类黄酮总量达到69.4 mg/g;实验所用剂量条件下,黑青稞籽皮类黄酮表现出了对三种缺氧损伤细胞的保护作用,并能有效清除活性氧自由基,提高过氧化物歧化酶的活性。结论黑青稞籽皮提取物发挥缺氧损伤保护作用的物质基础可能是花青素类黄酮,它对防护活性氧介导的细胞损伤具有重要意义。 Objective To investigate the protective effects of black barley pericarp flavonoids against hypoxia in 3 hypoxia-induced cell injury models. Methods The flavonoids were extractedfrom black barley pericarp using citric acidalcohol maceration method. The flavonoid content was assayed by two methods and the protective effects against hypoxia were verified by 3 cell models. Results 30.3 mg pericarp extracts per 1 g dry black barley were obtained. The spectroscopy and HPLC assays identified anthocyanidins were the main components offlavonoids. The extraction rate of anthocyanin was 68.6 mg/g, and delphinidin chloride represented 57 mg/g. The flavonoids were 69.4 mg/g of dry weight.The flavonoids present in acid-alcohol extract of black barley pericarp prevented cell from hypoxia injury by effectively eliminating reactive oxygenspecies(ROS) and increasing the activities of superoxide dismutase(SOD). Conclusion Theanthocyanin flavonoids extractedfrom black barley pericarp play a protective role in hypoxic injury, especially in controlling ROS-meditatedhypoxic cellular injury.
