宫颈癌中HPV感染与Rap1GAP基因及蛋白水平改变关系的研究 被引量:1

摘要 目的研究宫颈癌中HPV感染与Rap1GAP基因及蛋白水平改变的关系。方法以我院收治的70例宫颈癌患者为观察组,另选正常宫颈组织70例为对照组。分别对两组对象的宫颈组织进行DNA提取、总RNA提取、cDNA合成、PCR反应扩增。并检测HPV感染、Rap1GAP基因外显子E11和E19缺失率、Rap1GAP mRNA表达及Rap1GAP蛋白表达情况。结果观察组患者HPV感染明显高于对照组(P<0.05)。观察组HPV阳性和阴性组E11和E19的缺失率比较,差异无统计学意义(P>0.05)。Rap1GAP mRNA、Rap1GAP蛋白在观察组HPV阳性组组织中的表达率、阳性表达率明显低于HPV阴性组(P<0.05)。宫颈癌中HPV感染与Rap1GAP mRNA的表达呈正相关(r=0.578),与Rap1GAP蛋白的阳性表达呈正相关(r=0.554)。结论宫颈癌中HPV感染与Rap1GAP mRNA的表达呈正相关,与Rap1GAP蛋白的阳性表达呈正相关。 Objective To study the cervical HPV infection in relations with Rap1 GAP gene and protein level change.Methods A total of 70 cases of cervical cancer patients in our hospital were selected as the observation group,chose70 cases of normal cervical tissues as the control group. Respectively for 2 set of objects of cervical tissue DNA extraction, total RNA extraction, amplification cDNA synthesis, PCR reaction. And detection of HPV infection, Rap1 GAP gene exon E11 and E19 loss rate, Rap1 GAP Rap1GAP mRNA expression and protein expression. Results The observation group of patients HPV infection was significantly higher than control group(P〈0.05). HPV positive and negative E11 and the E19 lack of rate comrpared in the observation group was no statistically significant difference(P〉0.05).In the observaition group, the HPV positive group organization, Rap1 GAPmRNA and Rap1 GAP protein expression rate and positive expression rate was significantly lower than the HPV negative group(P〈0.05). Cervical HPV infection and Rap1 GAP mRNA expression were positively correlated(r = 0.578), and Rap1 GAP protein positive expression were positively correlated(r = 0.554). Conclusion Cervical HPV infection and Rap1 GAP mRNA expression were positively correlated, and Rap1 GAP protein positive expression were positively correlated.
