海参花水提物对环磷酰胺诱导小鼠生殖系统受损的保护作用 被引量:8

Protective Effect of Water Extracts of Sea Cucumber Ovum on Reproductive System of Premature Ovarian Failure Mice

摘要 目的探讨海参花水提物对环磷酰胺诱导的小鼠生殖系统受损、卵巢早衰的保护作用。方法昆明♀小鼠随机分为空白组、模型组、阳性对照组(戊酸雌二醇0.085 mg·kg-1)、海参花水提物低、中、高剂量组(分别为150,300,600 mg·kg-1),除空白组每天注射生理盐水外,其余各组连续5 d腹腔注射环磷酰胺28 mg·kg-1,造成小鼠生殖系统紊乱,建立小鼠卵巢早衰模型,并同时灌胃给予相应药物。放免法检测血清雌二醇含量(E2)、孕酮(P)含量,比较各组小鼠间子宫、卵巢、脾、胸腺等脏器重量,并进行小鼠卵巢、子宫组织形态学观察。结果连续用药4周后,与模型组相比,海参花水提物各剂量组均能显著增加小鼠的子宫、卵巢、胸腺等脏器重量(P<0.05或P<0.01),显著提高E2、P含量(P<0.05或P<0.01),增加子宫内膜平均厚度(P<0.01),改善卵巢形态。结论海参花水提物可增加子宫内膜厚度,对卵巢早衰发挥防治作用,对磁性小鼠生殖系统有一定的保护作用。 OBJECTIVE To investigate the protective effects of water extracts of sea cucumber ovum on deficiency of the genital system injured by cyclophosphamide. METHODS Female Kunming mice were selected and randomly divided into 6 groups included control group, model group, positive group (estradiol valerate 0.85 mg·kg-1), and low-, middle-, high-dose sea cucumber ovum group(150, 300, 600 mg·-1) with 10 animals in each group, except the control group injected with normal saline, others were injected with cyclophosphamide(28 mg18a·-1) daily for successive 5 d to induce reproductive system disorders and premature ovarian failure in mice. Detected the level of estradiol(E2) and progesterone(P) by radioimmunoassay, all mice were compared the weight of organs, such as uterus, ovarian, spleen, and thymus, observed for the form of uterus and ovarian. RESULTS Sea cucumber ovum could increase the weight of organs, such as uterus, ovarian, and thymus gland of mice obviously(P〈0.05, P〈0.01), significantly improved the level of E2 and P(P〈0.05, P〈0.01), increased the thickness of endometrial (P〈0.01), improved the form of ovarian obviously. CONCLUSION Sea cucumber ovum can increase the thickness of endometrial, prevent the premature ovarian failure, promote and protect the reproductive system of female mice.
