摘要
目的:测定蒙药消肿九味散中大黄酸、大黄素、大黄酚3种活性成分的含量。方法:采用高效液相色谱(HPLC)法,Dimaonsil-C18(250 mm×4.6 mm,5μm)色谱柱;以甲醇-0.1%磷酸溶液(85∶15)为流动相;流速:1.0 mL·min-1;检测波长:254 nm,柱温为25℃。结果:大黄酸、大黄素、大黄酚3种成分分别在0.101 8~0.508 8μg、0.117 0~0.585 0μg、0.256 0~1.280 0μg范围内与其峰面积呈良好的线性关系,相关系数均大于0.9997;且样品中3种成分的平均加样回收率(n=6)分别为98.98%、99.54%、97.36%,RSD均小于1.32%。结论:建立的含量测定方法简便、快速、灵敏、准确,可以从该制剂中原料药材君药的角度用于该制剂的质量控制。
Objective : To determinate the contents of three active ingredients in Mongolian medicine of Xiaozhong Jiuwei Powder, including rheinic acid, rheum emodin and chrysophanol. Methods:High Pressure Liquid Chromatography(HPLC) was adopted, using the Dimaonsil - C18 (250mm × 4.6mm, 5μm) as chromatographic column with 25 ℃ , methanol - 0. 1% phosphoric acid solution(85:15)as mobile phase. The flow rate was 1.0 mLrain-1 and detection wavelength was 254 nm. Results:The three ingredients of rheinic acid, rheum emodin and chrysophanol showed good linear relation with their peak areas within the limits of 0. 1018-0.5088μg( r = 0. 9997 ) ,0.1170 - 0. 5850 μg( r = 0. 9997 ) and 0. 2560 - 1. 2800 μg( r = 0. 9998 ) , respectively. All their correlation coefficients were greater than 0.9997. The mean recovery of 98.45% ( 1.32% ) for rheinic acid,99.60% ( 1.09 % )for rheum emodin and 97.36 % ( 1.17 % )for c hrysophanol were obtained ( n = 6), and all the RSD values were less than 1.32%. Conclusion:The method established in this study is simple,high accuracy,precision and specific,and thus it can be used to control the quality of preparation perspective from monarch drug in a crude medicinal materials of a prescription.
