小鼠腹腔动—静脉瘘模型构建及中药复方SP的干预 被引量:1

摘要 目的:建立小鼠腹腔动-静脉瘘模型及中药复方SP(30 mg· kg^-1,丹酚酸B∶丹皮酚=3∶1)对其影响。方法4号针头刺穿下腔静脉-腹主动脉联合壁造瘘,2 w后进行病理形态学分析,另一组动物造模后给予相应药物,连续2 w,全自动生化分析仪测定血清中乳酸脱氢酶(LDH)以及肌酸激酶(CK)的含量,计算全心重/体重和左室重/体重比,对左心室进行HE染色。结果造模4 w后,模型组全心重/体重比、左室重/体重比及血清中LDH、CK活性增高( P<0.01),病理形态学检查显示该模型复制成功;复方SP组全心重/体重比、左室重/体重比及血清中LDH、CK活性降低( P<0.01~0.05)。结论小鼠腹腔动-静脉模型复制成功,中药复方SP对小鼠腹腔动-静脉瘘导致的心肌肥厚有改善作用。 Objective To establish a model of arteriovenous fistula in mice and to evaluate the treatment of the compound SP( 30 mg · kg ^- 1 , salvianolic acid B: paeonol = 3:1 ). Methods We created the fistula by using the size 4 needle piercing the abdominal aorta and getting into the inferior vena cava. Pathological analysis was made 2 weeks later. The other group was given some other medicines for two weeks after the establishment of the model. Two weeks after treatment, LDH and CK in serum were determined, the ratios of heart weight to body weight and left ventricular weight to body weight were calculated, HE staining of the left ventricle was taken for the pathology. Results In the model group, the ratios of heart weight to body weight and the left ventricular weight to body weight and the LDH, CK levels in serum were all higher than those in the control group (P 〈 0.01 ). Pathological analysis indicated the successfully reproduced model. The ratios of heart weight to body weight,left ventrieular weight to body weight and the CK, LDH levels in the serum of the compound SP group compared with the control group were obviously decreased ( P 〈 0.01 - 0.05 ). Conclusions The model of arteriovenous fistula in mice reproduced successfully, and compound SP improved the myocardial hypertrophy caused by arteriovenous fistula in mice.
