慢性阻塞性肺疾病稳定期大鼠mtDNA ATPase 6亚基基因的变化 被引量:1

摘要 目的观察慢性阻塞性肺疾病(COPD)稳定期大鼠mtDNA ATPase 6亚基基因的改变。方法将20只SD大鼠按照宋一平等介绍的方法(香烟雾熏加脂多糖气管滴入法)复制大鼠COPD模型后提取mtDNA,采用PCR技术扩增后测序,检测突变点。结果 SD大鼠mtDNAATPase 6亚基基因存在多态性,COPD稳定期模型大鼠存在一定数量点突变,mtDNA ATPase 6:A8000T、G8074A、A8439G、C8009T、C8062。结论 COPD稳定期大鼠mtDNAATPase 6亚基基因会发生突变,由此可导致所编码的氨基酸的改变,有可能是酶活性改变的原因。 Objective To observe the genetic changes of mtDNA ATPase6 of rats with chronic obstructive pulmonary disease in stabilization period. Methods 20 SD rats were used to copy COPD model according to Song Yiping's methods (tracheal instillation with fumigation and lipopolysaccharide). MtDNAs were extracted and PCR amplification was used to detect point mutations. Results The subunit genes of mtDNAAT Pase6 of SD rats have the nature of polymorphism. The rats with CPOD in stabilization period have a certain amount of point mutations, such as mtDNAATPase 6: AS000T, G8074A, A8439G, C8009T, C8062. Conclusion The subunit genes of mtDNAATPase 6 COPD of rats in stabilization period have mutations, which can lead to the change of coding amino acid. It is probably the reason for the changes of enzyme activity.
