癌基因PIM-1通过调控mir-17-5p表达调控鼻咽癌细胞的转移 被引量:1

摘要 目的:探讨PIM-1蛋白参与鼻咽癌细胞转移的机制。方法采用shRNA将CNE-2Z鼻咽癌细胞中的PIM-1的表达进行沉默,并采用realtime对mir-17-5p的表达进行检测;采用shRNA将CNE-2Z鼻咽癌细胞中的mir-17-5p的表达进行沉默,采用transwell实验比较mir-17-5p转染前后sh1-PIM-1细胞的迁移能力。结果 Realtime及Western Blot结果显示PIM-1沉默后mir-17-5p的表达水平显著升高。transwell结果显示,mir-17-5p的过表达增强CNE-2Z细胞的迁移能力。结论癌基因PIM-1通过调控mir-17-5p的表达,可以调控鼻咽癌肿瘤细胞的转移。 Objectives The purpose of this study was to investigate. Methods PIM 1 expression in CNE-2Z cell line was knocked down by shRNA,the Silence efficiency was evaluated by realtime PCR and Western Blot.The migration of nasopharyngeal carcinoma was analyzed by transwell assay. Results We found that the expression of mir-17-5p was upregulated after PIM1 knock down.Meanwhile,the migration ability was upregulated after mir-17-5p wastransfected. Conclusion PIM - 1 can promote the metastasis of nasopharyngeal carcinoma through the regulation of mir-17-5 p expression regulation.
