摘要
应用荧光探针、紫外光谱技术和黏度法研究了荭草苷和DNA分子间的相互作用.研究结果表明,在pH为7.4的Tris-HCl缓冲体系中,荭草苷可将盐酸小檗碱从DNA双螺旋中挤出,导致其荧光猝灭.根据热力学参数确定了荭草苷与DNA主要以疏水作用力结合;25℃和40℃两个不同温度下荭草苷与DNA之间的结合常数分别为1.541×103 L·mol-1和1.602×103 L·mol-1,结合位点数分别为0.7888和0.7242;黏度实验进一步证明荭草苷是以嵌入结合的方式与DNA结合.
The interaction of orientin and DNA was studied using fluorescent probe,UVspectrometry and viscosity method.The results showed that orientin can extrude berberine hydrochloride(BR) from the DNA double helix and cause the decrease of the fluorescence intensity.The main binding force between orientin and DNA is hydrophobic force according to thermodynamic parameters.The binding constant K at 25℃ and 40℃ were calculated as 1.541×10^3 and 1.602× 10^3 L · mol^-1,and the binding sites number n as 0.7888 and 0.7242,respectively.The viscosity experiments further proved that orientin binds with DNA in a mode of intercalative binding.
