摘要
目的观察丁桂儿脐贴、丁桂儿口服及联合用药对脾胃虚寒幼龄大鼠模型的影响。方法将136只SD大鼠按随机数字表法分为11组(每组12或13只):正常对照组,模型对照组,模型赋形剂组,丁桂儿脐贴小、中、大剂量组,香砂六君子丸+丁桂儿脐贴小、中、大剂量组,香砂六君子丸组,丁桂儿口服组。采用食醋法制备脾胃虚寒动物模型,观测各组大鼠体质量、食量、胃液游离胃酸及总酸度、胃蛋白酶活性、胃排空率及小肠推进率;采用ELISA法检测大鼠血浆八肽胆囊收缩素(CCK-8)及胃动素(MTL)含量。结果与正常对照组比较,模型大鼠体质量逐渐降低,小肠推进率明显降低,MTL分泌显著减少,而胃酸及胃蛋白酶分泌无显著变化,提示模型主要反映胃肠动力不足。建模后14d,与模型对照组比较,除丁桂儿脐贴中剂量组大鼠体质量有改善外,含丁桂儿的其他各组大鼠体质量无明显改变(P>0.05);含丁桂儿的各组大鼠胃排空率及小肠推进率无明显变化(P>0.05)。与模型对照组比较,香砂六君子丸+丁桂儿脐贴中剂量组血浆CCK-8含量显著降低(P<0.01);与正常对照组比较,模型对照组及模型赋形剂组大鼠血浆MTL含量显著降低(P<0.05),丁桂儿脐贴小剂量组大鼠血浆MTL含量显著升高(P<0.05)。结论丁桂儿脐贴及联合用药可参与调节胃肠激素的分泌,并改善脾胃虚寒动物模型的症状。
Objective To observe the influences of Dingguier umbilical paste,oral Dingguier and combined medication on spleen-stomach deficiency-cold in j uvenile rats.Methods A total of 1 3 6 SD rats were randomly divided into 1 1 groups:normal control group,model control group, model vehicle group,small-dose Dingguier umbilical paste group,medium-dose Dingguier umbili-cal paste group,large-dose Dingguier umbilical paste group,Xiangshaliuj un pill+small-dose Ding-guier umbilical paste group,Xiangshaliuj un pill+medium-dose Dingguier umbilical paste group, Xiangshaliuj un pill+large-dose Dingguier umbilical paste group,Xiangshaliuj un pill group,and o-ral Dingguier group,with 1 2 or 1 3 rats in each group.Spleen-stomach deficiency-cold was induced by vinegar feeding.Body weight,food intake,gastric acid secretion,gastric acidity,pepsin activity,gastric emptying rate and small intestinal propulsion rate were measured in all the 1 1 groups.In addition,plasma levels of cholecystokinin octapeptide (CCK-8 )and motilin (MTL)were deter-mined by ELISA.Results Compared with normal control group,body weight,small intestinal propulsion rate and MTL levels were decreased but gastric acid and pepsin secretion was not changed in model control group,suggesting that rats with spleen-stomach deficiency-cold had gas-trointestinal motility disorders.Compared with model control group,body weight was improved in medium-dose Dingguier umbilical paste group,and plasma CCK-8 levels were decreased in Xiang-shaliujun pill+medium-dose Dingguier umbilical paste group 14 days after model establishment (P〈0.01).However,compared with model control group,body weight in other groups and gas-tric emptying rate and small intestinal propulsion rate in rats treated with Dingguier were not changed significantly (P〉0.05).Compared with normal control group,plasma MTL levels de-creased in model control group and model vehicle group,but increased in small-dose Dingguier umbilical paste group (P〈0.05).Conclusion Dingguier umbilical paste and combined medication can regulate the secretion of gastrointestinal hormones and improve the symptoms of spleen-stom-ach deficiency-cold in j uvenile rats.
