小鼠急性心肌梗死模型建立及左心室重构评价 被引量:6

Establishment of mouse model with acute myocardial infarction and evaluation of left ventricular remodeling

摘要 目的探讨小鼠急性心肌梗死模型建立及左心室重构评价。方法取40只C57B/L6雄性小鼠,30只结扎冠状动脉(冠脉)左前降支,建立急性心肌梗死模型(A组),10只予假手术为对照(B组)。术前1d和术后4周心脏超声评价左心室舒张末内径(LVEDD)和左室射血分数(LVEF)。术后4周测量小鼠心脏重量/体重(HW/BW)比值,取心脏标本做Masson染色测量梗死纤维化面积,WGA染色测量小鼠心肌细胞横截面积。结果 A组心肌梗死模型建立成功率为73.3%(22/30)。A组小鼠术后4周LVEDD、心肌细胞横截面积和HW/BW比值均大于B组[(4.45±0.57)mm vs.(3.44±0.36)mm、(746.60±74.48)μm2 vs.(296.20±37.21)μm2和0.0065±0.0010vs.0.0054±0.0004](P<0.05)。A组小鼠术后4周LVEF低于B组[(46.95±6.72)%vs.(73.79±6.56)%](P<0.05)。A组平均梗死后纤维化面积为(48±11)%。结论采用结扎冠脉前降支可成功建立小鼠急性心肌梗死模型。LVEDD、LVEF HW/BW比值等可用于综合评价梗死后心肌重构程度。 Objective To establish the mouse model with acute myocardial infarction(AMI)and evaluate left ventricular(LV)remodeling.Methods Left anterior descending coronary arteries were ligated in 30C57B/L6male mice to establish AMI model(group A)and 10C57B/L6male mice were taken as sham operation controls(group B).LV end diastolic diameter(LVEDD)and LV ejection fraction(LVEF)were evaluated by cardiac ultrasound one day before operation and in the fourth week after operation.Heart weight/body weight(HW/BW)ratio was measured in the fourth week after operation.Infarction fibrosis area and myocyte cross-sectional area were measured by Masson staining and WGA staining,respectively.Results AMI models were established successfully in 73.3% mice in group A.In the fourth week after operation,LVEDD,myocyte cross-sectional area and HW/BW ratio were greater in group A than those in group B[(4.45±0.57)mm vs.(3.44±0.36)mm,(746.60±74.48)μm^2 vs.(296.20±37.21)μm^2 and 0.0065±0.0010vs.0.0054±0.0004](P〈0.05).But LVEF was lower in group A than that in group B[(46.95±6.72)% vs.(73.79±6.56)%](P〈0.05).The infarction fibrosis area in average was(48±11)%in group A.Conclusion AMI model in mice can be established successfully by ligating the left anterior descending coronary arteries.The indicators of LVEDD,myocyte cross-sectional area and HW/BW ratio can be used for comprehensively evaluating the severity of LV remodeling after AMI.
