摘要
目的探讨3’3-二吲哚甲烷(DIM)联合化疗药物5-氟尿嘧啶(5-FU)对胃癌细胞增殖的影响。方法 MTT法检测DIM、5-FU单药及联用对不同分化程度胃癌细胞株(MKN28、SGC7901、AGS、MKN45、HGC27)增殖的影响,计算药物对不同胃癌细胞的半数抑制浓度IC50及联用效应。结果 DIM及5-FU单药对不同分化程度胃癌细胞增殖的半数抑制浓度(IC50)范围分别为23~42μmol/L及9.5~95.9μmol/L,两者联用对胃癌细胞的抑制率较单药高,但仅对中分化胃癌细胞株SGC7901和低分化胃癌细胞株MKN45的联用指数(CI)均介于0和1之间。结论 DIM及5-FU单药均能抑制体外培养的不同分化程度胃癌细胞增殖,两者联用对抑制中分化胃癌细胞株SGC7901和低分化胃癌细胞株MKN45的增殖具有协同效应。
Objective To observe the in vitro inhibitory effects of 3,3'-Diindolylmethane(DIM) plus 5-fluorouracil(5-FU) on various gastric cancer cell lines(MKN28, SGC7901, AGS, MKN45, HGC27). Methods The inhibitory effects of DIM and 5-FU either in combination or alone were measured by MTT assay and the synergistic effect of the two drugs was evaluated by calcusyn software according to the median-effect principle. Results The IC50 of DIM and 5-FU on all the gastric cancer cell lines were ranged from 23 to 42 μmol / L and 9.5 to 95.9 μmol / L, respectively. The combination of DIM and 5-FU showed an accelerated inhibitory effect,but the two agents were synergistic only when used concurrently on gastric cancer cell line SGC7901 and MKN45. Conclusions These results indicated that both DIM and 5-FU showed dose-dependent inhibitory effect on gastric cancer cell lines(MKN28, SGC7901, AGS, MKN45, HGC27), and the combination of them was synergistic on gastric cancer cell line SGC7901 and MKN45.
