摘要
目的:研究白花蛇舌草多糖对游泳小鼠的抗疲劳作用及其体外抗氧化活性。方法:将白花蛇舌草多糖溶液3个剂量(0.5、1.0、2.0 g/kg)给小鼠灌胃,连续10 d。末次给药1 h后,除空白对照组外,其他组小鼠负重游泳,记录小鼠力竭游泳时间,取血分离血清,取肝脏和肌肉制备组织匀浆。采用硫代巴比妥酸法测定血清丙二醛(MDA)含量,采用黄嘌呤氧化酶法测定血清超氧化物歧化酶(SOD)含量,采用蒽酮-硫酸法测定糖原含量。实验还采用邻苯三酚自氧化法观察白花蛇舌草多糖的清除氧自由基能力,采用Fenton反应法测定清除羟自由基能力。结果:白花蛇舌草多糖能显著延长游泳小鼠的力竭游泳时间,提高肝糖原和肌糖原贮备,降低血清MDA水平,提高血清SOD水平。白花蛇舌草多糖还在体外具有良好的清除氧自由基和羟自由基活性。结论:白花蛇舌草多糖具有体外抗氧化活性,并可延缓疲劳的发生,这一作用可能与其抑制脂质过氧化损伤有关。
Objective: Anti-fatigue effects on the swimming mice and antioxidation in vitro of Hedyotis diffusa polysaccharides were studied. Methods: Hedyotis diffusa polysaccharides (0.5, 1.0, 2.0 g/kg) were given to mice for 10 days. 1 h after the last administration, mice were loaded to swim except the blank control group and the exhausted swimming time was recorded. Blood serum were separated and liver and muscle tissue homogenates were prepared. Serum malondialdehyde (MDA) was determined by thiobarbituric acid method. Serum superoxide dismutase (SOD) was measured by xanthine oxidase method. Glycogen content was determined by anthrone-sulfuric acid method. Oxygen free radical scavenging ability of Hedyotis diffusa polysaccharides was observed by pyrogallol autoxidation method and hydroxyl radical scavenging ability by Fenton reaction. Results: Hedyotis diffusa polysaccharides can significantly prolong the exhausted swimming time in swimming mice, increase liver glycogen and muscle glycogen reserves, lower serum MDA levels and increase serum levels of SOD. Hedyotis diffusa polysaccharides exhibited excellent scavenging oxygen radicals and hydroxyl radicals in vitro. Conclusion: Hedyotis diffusa polysaccharides has antioxidation effects in vitro and delay the occurrence of fatigue. This effect may be related to its inhibition effect on lipid peroxidation.
