摘要
目的观察纺锤体检查点蛋白(Cenp-E)在肝癌组织和人类正常肝组织表达水平的差异,以探讨Cenp-E蛋白在肝癌细胞中的表达及临床意义。方法用荧光定量PCR和Western blot分别检测Cenp-E基因和蛋白在肝癌组织和正常肝组织的表达水平。结果正常肝组织中Cenp-E基因mRNA水平(0.023 78±0.009 73)明显高于肝癌组织(0.014 58±0.008 73);Cenp-E蛋白表达量在正常肝组织中(0.656±0.012)明显高于在肝癌组织中的表达(0.301±0.009),且二者有显著性差异。结论 Cenp-E低表达在肝癌的发生过程中起重要作用。
Objective To study the differential expression and its clinical significance of the spindle checkpoint protein Cenp-E gene between the human hepatoma and hepatic tissue. Methods Real-time fluorescent-quantitation PCR and Western blot were employed to detect the expressions of the Cenp-E mRNA and protein, respectively. Results The expression of the Cenp-E mRNA in human hepatic tissue(0. 023 78 ± 0.009 73 ) was higher than that in human hepatoma tissue(0.014 58 ±0.008 73). The expression of the Cenp-E protein in human hepatic tissue(0. 656 ±0.012) was higher than that in human hepatoma tissue(0. 301 ± 0.009). Conclusion Cenp-E gene low expression in human hepatocellular carcinoma plays an important role in the its developmental process.
