Caspase-9在乳腺浸润性导管癌组织中的表达及其临床意义

摘要 目的:探讨半胱氨酸天门冬氨酸特异的蛋白酶9(Caspase-9)在乳腺浸润性导管癌组织中的表达及其与临床病理因素的关系。方法:采用免疫组织化学SP方法研究38例乳腺浸润性导管癌(breast infiltrating ductal carcinoma,IDC)、32例乳腺纤维腺瘤及其肿物旁的正常乳腺组织中Caspase-9的表达水平,观察评估Caspase-9的阳性表达与患者年龄、肿瘤大小、淋巴结转移等临床病理因素的相关性,并探讨与乳腺浸润性导管癌的临床分期、组织学分级的关系。结果:乳腺浸润性导管癌、乳腺纤维腺瘤组织中的Caspase-9的阳性表达均低于正常乳腺组织(P<0.05),且乳腺浸润性导管癌明显低度表达(P<0.01)。其表达水平与患者年龄、肿瘤大小、淋巴结转移等不相关(P>0.05),与临床分期、组织学分级明显负相关(P<0.01)。结论:Caspase-9低表达与乳腺浸润性导管癌的恶化显著相关,该指标有助于乳腺浸润性导管癌的预后判断,其可能成为潜在乳腺肿瘤标志物,并成为治疗的靶点。 Objective: To investigate the expression of Caspase-9 protein in breast infiltrating ductal carcinoma( IDC) and its clinical significance. Methods: The expression of Caspase-9 was detected by streptavidin-perosidase( SP) with tissues of thirty-eight cases of IDC and thirty-two cases of fibroadenoma,including normal tissues adjacent to tumor. Positive expression of Caspase-9 relation with clinicopathological factors such as age,tumor size,lymph node metastasis were observed. Furthermore,clinical stage and histopathologic grade were explored in IDC. Results: The expression of Caspase-9in IDC tissues and fibroadenoma tissues were lower than that in adjacent normal tissues( P〈0. 05). Meanwhile,the IDC tissues was significantly down-regulated( P〈0. 01). The expression of Caspase-9 was independent of clinicopathological parameters,such as age,tumor size,lymph node metastasis( P〉0. 05). However,It had obvious negative correlation with clinical stage and histopathologic grade( P〈0. 01). Conclusion: The lower expression of Caspase-9 closely correlates to the progress of breast infiltrating ductal carcinoma. The index greatly contributes to judgment of prognosis of breast infiltrating ductal carcinoma. Indicating that Caspase-9 might be used as a tumor biomarker and a potential therapeutic target for inhibiting the invasive of breast infiltrating ductal carcinoma.
