摘要
为给山羊品种的选育和种质鉴定提供理论依据,以黔北麻羊为试验对象,采用DNA池结合PCR产物直接测序及PCR-SSCP技术对GHSR基因进行单核苷酸多态性检测。结果表明:在黔北麻羊GHSR基因外显子2处检测到1个同义位点G200A,氨基酸由甘氨酸变为丝氨酸,表现为3种基因型,分别命名为GG、GA和AA,其频率分别为0.507 4、0.408 0和0.084 6,等位基因G、A频率分别为0.711 4和0.288 6。χ2检验表明,黔北麻羊GHSR基因G200A位点分布符合Hardy-Weinberg平衡(P>0.05)。基因型与体重体尺的关联分析显示,GG和GA基因型个体的体重显著高于AA基因型个体,胸深显著小于AA基因型个体(P<0.05),而其余5个指标差异不显著(P>0.05)。
In order to provide a theoretical basis for variety breeding and germplasm identification,Qianbei Ma Goat was chosen as testing subject,the DNA pooling was constructed,the technology of direct sequencing of PCR products and PCR-SSCP were used to detect the single nucleotide polymorphism of GHSR gene.The results showed that G200A was detected in exon 2 of GHSR gene,the SNP(G200A) was a sense mutation,which made Gly to Ser and was devided into three genotypes:GG,GA and AA.The frequencies of these genotype were 0.5074,0.4080 and 0.0846,respectively.The frequency of allele G and A were 0.7114 and 0.2886.x2 test indicated that this polymorphism of G200A of GHSR gene fit with Hardy-Weinberg equilibrium in QianBei Ma goat (P> 0.05).The study of relevance analysis between genotype and body weight and size revealed that the body weight of individuals with genotype GG and GA were significantly higher than that of individuals with genotype AA,and smaller than those for chest depth (P<0.05),but no significant difference in the others traits (P>0.05).
