丹参多酚酸盐介导的心梗后大鼠心功能和心室重构的改善 被引量:4

Therapeutic injection of Depside Salt From Salvia Miltiorrhiza induces myocardial remodeling and enhances cardiac function of ischemic heart failure

摘要 目的 探讨丹参多酚酸盐对心梗后大鼠心功能和心室重构的影响.方法 结扎大鼠冠状动脉左前降支建立心梗模型.术后6周内治疗组肌肉注射丹参多酚酸盐1 mg/kg;非治疗组注射生理盐水.6周后心导管法测定大鼠心功能,马松染色了解心肌组织胶原含量,免疫组化了解梗死周围区新生血管密度.结果 治疗组大鼠心功能好转显著优于对照组[max dp/dt (4788.2+1069.3)mm Hg/s比(3325.2+775.9)mm Hg/s,P<0.01],治疗组梗死边缘区的血管密度明显高于对照组[(98.7+12.9/高倍镜视野比69+11.0/高倍镜视野,P<0.01],治疗组组织胶原含量明显低于对照组[(25.8+5.9)%比(35.1+9.1)%,P<0.01].结论 丹参多酚酸盐可以改善大鼠心梗后心功能,降低梗死区胶原含量,促进血管新生,改善心室重构. Objective To study the effects of injection of Depside Salt From Salvia Miltiorrhiza on car diac function of myocardial infarction rat.Methods Acute myocardial infarction model was built by left anterior decending coronary artery ligation.Lewis rats underwent left anterior descending coronary artery ligation followed by intramuscular injection of Depside Salt From Salvia Miltiorrhiza (n=8) or Saline (n=8) for six weeks.Cardiac hemodynamics parameters were done with pressure catheter.Myocardial histology was analyzed by immunohistochemistry staining.Results The hemodynamicsmanifested improvedin treatment animals(P<0.01).The bordersone vessels densityin treatment group were demonstrated increased (P<0.01),as well as less collagen volume fraction (P<0.01).Conclusion Depside Salt From Salvia Miltiorrhiza improves cardiac function in myocardial infarction rat,mediates the effect of ameliorating the vessel regeneration and the collagen content.
