摘要
目的:建立高效液相色谱法同时测定五加芪菊颗粒中刺五加(以紫丁香苷计)和菊花(以3,5-O-二咖啡酰基奎宁酸计)含量的方法,为相关研究提供方法学参考。方法采用DiamonsilC18色谱柱(250 mm×4.6 mm,5μm),流动相:甲醇为流动相A,0.1%磷酸溶液为流动相B,进行梯度洗脱。检测波长为348 nm。结果紫丁香苷和3,5-O-二咖啡酰基奎宁酸的线性范围分别为0.10~40.0μg/mL(r=0.9984)、0.05~10.0μg/mL(r=0.9991),方法回收率分别为98.76%(n=6)、94.26%(n=6)。结论所建立的方法准确、灵敏度高、重现性好,在同一试验过程中,同时测定五加芪菊颗粒中刺五加、菊花含量。
ObjectiveTo establish a method for simultaneously determining the content of acanthopanax (as measured by syringin), Chrysanthemum(as measure by (-)-3,5-Dicaffeoyl quinic acid) in Wujia Qiju granule.MethodsSimultaneously determined by HPLC on DiamonsilC18 (250 mm×4.6 mm,5 μm) by mobile phase of methanol (A) and 0.1% phosphonic acid solution (B) with gradient elution. Detection wavelength is 348 nm.ResultsThe linear concentration ranges of the calibration curves for syringin and (-)-3,5-dicaffeoyl quinic acid were 0.10~40.0 μg/mL(r=0.9984),0.05~10.0 μg/mL(r=0.9991), respectively. The average recoveries for syringin and (-)-3,5-dicaffeoyl quinic acid were 95.42% (n=6),91.73%(n=6), respectively.ConclusionThe method is accurate, highly sensitive and reproducible for simultaneously determination of content for acanthopanax and chrysanthemum in a single run by HPLC.
