心房颤动早期阶段心房肌细胞小电导钙激活钾通道的功能改变 被引量:2

摘要 目的观察小电导钙激活钾(SK)通道在心房颤动(AF)早期阶段的功能变化,并探讨其意义。方法成年比格犬分为快速心房起搏组(5只)和对照组(3只),快速心房起搏组犬经右心房快速起搏(400次/min)4 h,分离获取左心房肌细胞;对照组手术方法相同,但不进行起搏,获取左心房肌细胞;采用全细胞膜片钳技术,记录两组SK通道介导的钙激活钾电流(IK,Ca)电流密度变化。结果当钳制电位为-100 mV时,快速心房起搏组心房肌细胞电流密度为(-1.7±0.3)pA/pF,对照组为(-0.7±0.1)pA/pF,P<0.01;+80 mV时,快速心房起搏组心房肌细胞电流密度为(4.1±0.6)pA/pF,对照组为(-1.6±0.2)pA/pF,P<0.01。结论 SK通道的功能上调是AF初始阶段的重要分子机制之一。
