青蒿琥酯对糖尿病大鼠肾脏CTGF及TGF-β1表达的影响

Effects of artesunate on expression of CTGF and TGF-β1 in renal tissue of diabetic nephropathy rat model

目的:探讨青蒿琥酯(Art)对糖尿病大鼠肾脏结缔组织生长因子(CTGF)和转化生长因子-β1(TGF-β1)的影响。方法:雄性SD大鼠分为假手术组、糖尿病肾病模型组(DN模型组)、Art小剂量治疗组、Art大剂量治疗组。12周末观察大鼠尿微量白蛋白(mALB)、尿β2微球蛋白(β2-MG),免疫组化观察肾脏CTGF和TGF-β1的表达。结果:各组大鼠肾组织中CTGF和TGF-β1的表达:DN模型组和两组Art治疗组均显著高于假手术组(P<0.01),两组Art治疗组均低于DN模型组(P<0.01),Art大剂量治疗组表达低于小剂量治疗组(P<0.05)。结论:青蒿琥酯可抑制CTGF、TGF-β1在糖尿病肾病肾组织的表达,从而起到保护肾脏的作用。

Objective To investigate the expression of connective tissue growth factor （CTGF） and transforming growth factor-β1（TGF -β1） in renal tissue of diabetic nephropathy rat model treated with artesunate. Methods..40 male SD rats were randomly allocated into 4 groups, including sham-operated group treated with distilled water, diabetic nephropathy model group treated with distilled water, diabetic nephropathy group treated with low-dose of artesunate and diabetic nephropathy group treated with high-dose of artesunate. After 12 weeks the urine mALB, urine 132-MG, and the expression of CTGF and TGF-β1 in renal tissue were measured with immunohistochemical method. Results=The expression of CTGF and TGF-β1 was higher in renal tissue of the 3 groups of diabetic nephropathy groups than in the renal tissue of the sham-operated group（P〈0. 01）, it was lower in the 2 groups treated with artesunate than in the diabetic nephropathy model group（P〈0.01）, and it was lower in the group treated with high-dose of artesunate than in the low-dose of Artesunate group（P〈0.05）. Conclusion Artesunate inhibits the expression of CTGF and TGF -β1 in renal tissue of diabetic nephropathy ,which can protect the kidney.
