摘要
用寡核苷酸多态性基因分型(Spoligotyping)对2008年1月至2010年10月间采集的211株结核分枝杆菌临床分离株进行基因分型,然后对北京型结核分枝杆菌VNTR位点的多态性进行检测.选用的12个VNTR位点存在较高的基因多态性,该12位点组合分辨能力(HGI=0.99985)高于标准的VNTR-12位点组合(HGI=0.99546),其中多态性较高的7位点组合HGI指数为0.99977.因此,7位点组合可以作为四川地区北京家族分枝杆菌的一线分型方法.
Clinical M. tuberculosis isolates from Jan 2008 to Oct 2010 were genotyped by Spoligotyping, and Beijing family was further analyzed for the polymorphism of the twelve tandem repeat loci. Analysis based on MIRU-VNTR for 162 Beiiing isolates suggested that the selected 12 loci show a higher HGI value compared with the original 12 loci method. Using 7-locus VNTR of high polymorphism, the HGI value (0.9998) was obtained. It is suggested that the method of 7-locus can be used as the first-line mo- lecular typing tool for Beijing family isolates of M. tuberculosis.
