鱼精蛋白截短体在构建融合蛋白中应用的研究进展

摘要 融合蛋白在临床研究中的应用越来越广泛,尤其是在病毒感染和肿瘤的基因治疗方面,但是其自身也有诸多缺点。鱼精蛋白截短体(tP)因其相对分子质量小,具有DNA结合功能,编码序列寡核苷酸的设计与合成比较简单等优点,克服了融合蛋白欠稳定等缺点。因此,tP被用于融合蛋白的构建中,融合基因表达的融合蛋白已用于临床前研究,为病毒感染和肿瘤的靶向治疗提供了新思路。 Fusion proteins have been widely used in clinical researches,especially in viral infections and cancer gene therapy,but it also has many shortcomings.Truncated protamine is featured with small molecular weight,DNA binding function and relatively simple design and synthesis of oligo nucleotides coding sequence,which can overcome some shortcomings of fusion proteins,such as unstability.Thus,truncated protamine has been used for the construction of fusion proteins,which is expressed by fusion gene has been used in pre-clinical researches,providing a new approach for viral infections and targeted therapy of tumors.
