摘要
目的建立动物粪便中番茄红素的提取方法,以新疆地产番茄酱为原料,对大鼠进行灌胃试验,收集其粪便,对其中的番茄红素残留进行分析检测。方法采用正交分析方法,对液料比、提取时间、提取温度、提取次数等进行优化分析,并采用分光光度法对实验提取结果进行检测。结果以苏丹红Ⅰ色素为标准品,番茄红素提取的最佳工艺条件:液料比为4∶1(V/W),提取时间40min,提取温度35℃,提取次数3次,提取率>84%。建立回归方程Y=1.778 X+0.021 3,r2=0.998 7,符合检测要求。结论该方法对研究番茄红素在人体内的消化吸收具有一定的理论指导意义。
Objective To development the method of extraction of lycopene in the SD rats′excrement. Methods Through the orthogonal experiment and spectro photometry, the optimal conditions are obtained and the detection method is determined.Results The optimal extracting conditions and lycopene yields of extraction technology were described as follows:the best extracting solvent,ethyl acetate;the ratio of solvent to tomato sauce,4∶1(V/W);the extraction time,40 min;the extraction temperature of 35℃ and the extraction times,3 times.The standard curve exhibited a good linear relationship between absorbance and tony red Ⅰ concentration.Conclusion The method is perfect to the method on the study of the Lycopene in the human digestion and absorption,with certain theory and practical significance.
