摘要
从新城疫标准强毒株F48E9株克隆血凝素-神经氨酸酶(HN)基因,将HN基因转入昆虫杆状表达系统,转染昆虫细胞Sf9。经Western blot和间接免疫荧光鉴定,该系统成功表达了HN蛋白。重组HN蛋白单次免疫SPF鸡可诱导机体产生较高的抗体水平,为试验鸡提供较好的免疫保护。研究结果用于生产证明该重组HN蛋白可刺激机体产生较强的免疫反应,可用于生产有效的新城疫疫苗。
In this study,the haemagglutinin-neuramindase (HN)gene of F48E9 strain of Newcastle disease virus (NDV) was cloned and expressed in Sf9 insect cells by haculovirus expression system (Bae-to-Bac). The expression of recombinant HN protein was confirmed by western-blot and indirect immunofluorescenee assay. Robust antigen-specific antibody response and protective immunity in SPF chickens were induced by single immunization with recombinant HN protein. Their levels were similar to those induced by commercial inactivated ND vaccine. Taken together, this study demonstrated that the recombinant HN protein could stimulate strong immune responses, and could used as an effective vaccine candidate for ND.
