新疆维吾尔族妇女宫颈癌中P53、IGF-I、VEGF和Bcl-2、Bax的表达及临床意义 被引量:4

Expressions and clinical significance of P53,IGF-I,VEGF and Bcl-2,Bax in Uyghur women with cervical cancer

摘要 目的探讨P53、胰岛素样生长因子-I(insulin like growth factors,IGF-I)、血管内皮生长因子(vascular endothelial growth factor,VEGF)、Bcl-2和Bax在新疆维吾尔族妇女宫颈癌组织中的表达和临床意义。方法收集维吾尔族妇女浸润性宫颈鳞癌患者的组织36例为宫颈癌组,正常或者慢性宫颈炎宫颈组织32例作为正常对照组。分别运用实时荧光定量PCR(realtime fluores-cence quantitative PCR,RTFQ-PCR)和蛋白免疫印迹(western blot,WB)技术检测P53、IGF-I、VEGF、Bcl-2和Bax在不同宫颈组织中的mRNA和蛋白表达水平。结果RTFQ-PCR结果显示,与正常对照组相比,宫颈癌组患者宫颈组织中IGF-I、VEGF和Bcl-2表达均有不同程度的升高,P53、Bax表达降低,差异均有统计学意义(P<0.05);WB结果显示,与正常对照组相比,宫颈癌组患者宫颈组织中VEGF和Bcl-2的表达均有不同程度的升高,P53、Bax表达降低,差异均有统计学意义(P<0.05),而两组IGF-I蛋白的表达差异无统计学意义。结论 Bcl-2、IGF-I和VEGF在宫颈癌组织中过度表达,P53、Bax在宫颈癌患者宫颈组织中表达降低,提示其与宫颈癌的发生发展和浸润转移密切相关,为本地区宫颈癌的早期诊断、判断预后寻找有效的生物学指标。 Objective To investigate the expression of P53,IGF-I (insulin like growth factors),VEGF (vascular endothelial growth factor,VEGF),Bcl-2 and Bax with cervical cancer for clinical assessment. Methods Cervical tissue samples were collected from Uygur women with cervical lesions (invasive cervical cancer,ICC:36 cases,cervicitis:32 cases).The mRNA and protein expression levels of P53,IGF-I, VEGF,Bcl-2 and Bax in tissues from two groups were respectively examined by realtime fluorescence quantitative PCR (realtime fluores-cence quantitative PCR,RTFQ-PCR)and western blot (western blot, WB).Results The result of Q-PCR showed that compared with the control group,IGF-I,VEGF and Bcl-2 expression was increased in varying degrees of cervical cancer group,P53,Bax expression in cervical tissue to reduce cervical cancer,and the difference were statistically significant (P 〈0 05);The result of WB showed that compared with the control group,VEGF and Bcl-2 expression was increased in different de-grees.P53,Bax in cervical cancer patients in cervical cancer group tissues is reduced,and the differences were statistically significant (P 〈0.05).IGF-I protein difference between the two groups was not statistically significant.Conclusion The excessive expression of P53,IGF-I and VEGF in cervical cancer,decreasing expression of P53,Bax in patients' cervical tissue,suggesting close relation to the development and metastasis of cervical cancer,find effective biological prognostic indicators for early diagnosis of cervical cancer in this region.
