摘要
目的:评价 PREVL Isola 全自动微生物标本接种仪对常见标本的接种效果。方法用自动接种仪接种痰液标本与尿液标本各200份、咽拭子标本100份、支气管吸出物100份、血培养阳性标本100份(仪器法),同时用传统手工接种法(手工法)接种同样的标本,培养24~48 h 后观察菌落生长情况,比较接种仪接种与手工接种效果的差异。结果手工法和仪器法对700份标本培养24 h 后菌落生长阳性率分别为34.7%、40.4%,仪器法阳性率高于手工法(χ2=6.69,P <0.05)。仪器法培养阳性的痰液标本中有19份经鉴定认为有意义的菌株而手工法未培养出;仪器接种法培养24 h 后单个菌落较多,多数可直接上机鉴定,报告时间较手工法缩短24 h。尿培养阳性标本中有10份仪器法较手工法菌落计数高1个数量级。血培养阳性标本经接种仪初次接种培养阳性率为93%,而手工法为82%。咽拭子标本及支气管吸出物培养显示,仪器接种法除阳性率高于手工法外,单个菌落的分离效果亦优于手工法。结论PREVL Isola 全自动微生物标本接种仪可以提高培养的阳性率,优化单个菌落分离效果,缩短了检出时间,并可确保操作人员的安全。
Objective To evaluate the inoculation effect of PREVL Isola automatic inoculation system for common clinical specimens.Methods 200 sputum samples,200 urine specimens,100 throat swabs,100 bronchial aspirates and 100 blood culture positive specimens were inoculated by automatic inoculating apparatus.at the same the same specimens above were inoculated by traditional manual method.The growth of colonies was observed after 24 h to 48h of culture.The differences between inoculation effect were compared between inoculation system and manual method.Results After 24h culture the positive rate of 700 specimens was 34.7% (by manual method)and 40.4% (by instruments)respectively.The positive rate of instrumental method was higher than that of manual method (χ2 =6.69,P <0.05).19 significant strains from sputum samples inoculated by instrument were not detected by manual inoculation method.There were more single colonies after 24 h culture inoculated by Instrument method,and most of them can be i-dentified directly by machine,thus the report time could be shortened 24 h compared with manual method. The bacterial count of 10 strains inoculated by Instrument method was an order of magnitude higher com-pared with manual method among urine culture positive samples.The first subcultivation positive rate of instrumental inoculation method among positive blood culture samples was 93% compared to 82% for manual method.Throat swab specimens and bronchial aspirate culture showed that the separation effect of single colony inoculated by Instrument method was superior to the manual method besides the positive rate was higher than the manual method.Conclusion PREVL Isola automatic minoculation system can improve the positive rate of culture and optimize the single colony isolation.Meanwhile it can shorten the detection period and ensure the safety of operating personnel.
