摘要
目的建立以磺酸化-β-环糊精(S-β-CD)为手性选择剂的毛细管电泳法测定维拉帕米片中对映体含量。方法采用未涂壁熔融石英毛细管柱,背景电解质为30 mmol·L-1磷酸盐缓冲液,pH4.0,手性选择剂为S-β-CD。结果维拉帕米两对映体在质量浓度为0.04~0.20 g·L1内均呈现良好的线性关系(r〉0.999)。加样回收率为97.6%~102.8%,精密度小于2.2%。结论所建立的方法适于盐酸维拉帕米片中对映体含量测定。
Objective To develop a CE method for determination of enantiomers of verapamil hydrochloride tablets using randomly sulfated β-cyclodextrin.Methods The separation of the enantiomers was performed in a fused-silica capillary tube,in the buffer of 30 mmol·L-1NaH2PO4with S-β-CD as a chiral selector at pH4.0.Results This method was linear for enantiomers of verapamil in the range of 0.04-0.20 mg·mL-1 with correlation coefficients > 0.999.Accuracy was validated on the basis of quotient of the averaged measurements and the nominal value and it ranged from 97.6% to 102.8%,the RSDs were within 2.2%.Conclusions The method developed was proved to be simple,accurate and precision which can be used for the determination of enantiomers of verapamil hydrochloride tablets.
