摘要
目的探讨锌对鼠前列腺癌细胞RM-1侵袭能力和凋亡的影响。方法将RM-1细胞培养于含Zn2+的培养基中,Zn2+终浓度分别为0μmol/L(control)、100μmol/L、200μmol/L、250μmol/L、300μmol/L,继续培养24 h,MTT检测锌对RM-1细胞增殖的影响;RT-PCR检测肿瘤侵袭和凋亡相关基因mmp-2、mmp-9、bax和bcl-2 m RNA表达。结果 MTT检测结果显示,与对照组比较,不同浓度锌对RM-1细胞增殖抑制率明显增加(P<0.05);与100μmol/L组、200μmol/L组比较,250μmol/L组、300μmol/L组细胞增殖抑制率明显增加(P<0.01)。RT-PCR结果显示,与对照组比较,250μmol/L、300μmol/L组mmp-2和mmp-9 m RNA及bcl-2 m RNA表达水平明显降低(P<0.01);bax m RNA表达水平明显升高(P<0.01)。结论高锌可以降低RM-1细胞的增殖和侵袭能力,诱导RM-1细胞的凋亡。
Objective To investigate the effects of zinc on the invasion and apoptosis of RM-1 cells.Methods The RM-1 cells were exposed to different concentration of zinc for 24 h.MTT was used to detect the proliferation rate of RM-1cells.RT-PCR was used to detect the mRNA expression of invasion and apoptosis-related genes including mmp-2,mmp-9,bax and bcl-2.Results Compared with control group,the proliferation rates of RM-1 cells in different concentration of zinc groups were lower (P < 0.05).Compared with 100 μmol/L and 200 μmol/L groups,the proliferation rates of RM-1 cells in 250 μmol/L and 300 μmol/L groups were significantly inhibited (P < 0.01).Compared with control group,the expression of mmp-2,mmp-9 and bcl-2 mRNAs were decreased and the expression of bax mRNA was significantly increased in 250 μmol/L and 300 μmol/L groups (P < 0.01).Conclusion Zinc can inhibit proliferation and invasion,and induce apoptosis of RM-1 cells.
