动员大鼠自体骨髓干细胞修复损伤心肌 被引量:1

Mobilization of Autologous Bone Marrow Stem Cells to Repair Myocardial Injury in Rat

摘要 目的探讨应用rh G-CSF促进骨髓干细胞原位移植对损伤心肌的修复作用。方法采用异丙基肾上腺素腹腔注射建立大鼠急性心肌损伤模型;通过外源性细胞因子rh G-CSF动员骨髓干细胞增殖、释放、迁移归巢至损伤心肌局部;应用HE染色结合图像分析系统,对两组心肌进行对比性分析;应用八导记录仪检测心脏功能。结果原位移植组的心肌损伤面积小于对照组;原位移植组心脏收缩功能较对照组明显改善。结论腹腔注射异丙基肾上腺素可复制急性心肌损伤模型;应用骨髓干细胞动员剂rh G-CSF,可促进坏死心肌修复,提高心脏收缩功能。 Objective To investigate the repair effect for in-situ transplantation of bone marrow stem cells for acute myocardial injury in rats.Methods Acute myocardial injury model was established by intraperitoneal injection of isoproterenol 10 mg/kg in rat.Autologous bone marrow stem cells were mobilized by cytokine,released and homed to local injured myocardium.A comparative analysis on pathological section between two groups was performed by HE staining and image analysis system.Heart functions were determined by eight lead physiological polygraph system.Results The myocardial necrosis area in in-situ transplantation group was less than that of the control group.The cardiac systolic function was significantly improved in in-situ transplantation group compared to the control group.Conclusion Acute myocardial injury model could be established by isoprenaline administration.Myocardial necrosis were repaired and cardiac systolic function was improved by using cytokines.
