鞘内注射布美他尼在大鼠足底切口痛模型中的镇痛作用 被引量:2

ANALGESIC EFFECT OF INTRATHECAL INJECTION OF BUMETANIDE IN RATS WITH POSTOPERATIVE INCISIONAL PAIN

摘要 目的:研究鞘内注射钠钾氯联合转运蛋白1抑制剂布美他尼在足底切口痛模型大鼠的镇痛效应。方法:成年雄性SD大鼠36只随机分成对照组和布美他尼组,在切开足底前经鞘内注入人工脑脊液20μl或布美他尼100μg/20μl。在切开前1 d至切开后6 d分别进行疼痛累积评分、热痛阈及机械痛阈测定。结果:鞘内注射布美他尼降低大鼠切开后2 h至3 d的疼痛累积评分,增加伤口旁切开后2 h至6 d的热痛阈及切开后2 h至5 d的机械痛阈。结论:鞘内注射布美他尼可缓解足底切口痛模型大鼠的静息痛、热及机械性刺激的诱发痛,有希望成为新的术后镇痛方法。 Objective: To investigate the analgesic effect of intrathecal bumetanide(an inhibitor of sodiumpotassium-chloride co-transporter 1) in rats with incisional pain. Methods: Thirty six adult male SpragueDawley rats were randomly divided into control and bumetanide group. Bumetanide(100 μg/20 μl) or vehicle(20 μl) was intrathecally administrated prior to incision in the rats. Cumulative pain score, thermal paw withdrawal latency and mechanical withdrawal threshold were measured from the day before incision to the sixth day after incision. Results: Intrathecal injection bumetanide decreased cumulative pain scores from 2 h to 3 d following incision, increased thermal pain threshold 2 h to 6 d following incision, and mechanical pain threshold 2 h to 5 d following incision. Conclusion: Intrathecal bumetanide had analgesic effect on rest pain and evoked pain by thermal and mechanical stimulation in rats of incision pain model, which may serve as a promising therapeutic strategy for postoperative pain management.
